Khoo Si Toon Keng Tong Kongsi occupies a shophouse with an ornately decorated façe. As with nearby Boon San Tong Khoo Kongsi, it is an offshoot of the Khoo clan. While the Khoo clans trace their ancestry to a common progenitor established at Leong San Tong, Khoo Si Toon Keng Tong Kongsi was established by a branch of third-generation descendants.
Khoo Si Toon Keng Tong Kongsi itself is further split into five branches Yeoh Teik Tong Khoo Kongsi (Chneh Pang), Seow Tek Tong Khoo Kongsi (Cheng Pang), Sin Teik Tong Khoo Kongsi (Goh Pang), Swee Teik Tong Khoo Kongsi (Mooi Pang) and Swee Thong Tong Khoo Kongsi (Soo Pang).
